QMCF Technology - Mammalian Expression System
QMCF Technology is stable episomal expression system that uses mammalian cells and appropriate plasmids for expression of biologically active substances, such as recombinant proteins, recombinant antibodies, virus like particles (VLPs); and for expression of proteins of interest in cell-based assays of the putative (drug) substances.
QMCF plasmids contain mouse polyomavirus (Py) DNA replication origin which in combination with Epstein-Barr virus (EBV) EBNA-1 protein binding site as nuclear retention element ensure stable propagation of plasmids in the QMCF cells.
QMCF cell lines support stable maintenance and partitioning of QMCF plasmids by combination of two proteins EBV EBNA-1 and Py Large-T antigen that are expressed in QMCF cell lines. QMCF cell lines base on CHO, HEK293 and U2OS cell lines.
In comparison with transient protein production where plasmids get lost during cell division in few days, the QMCF plasmids stably replicate in QMCF cells and maintain their copy number. Using QMCF Technology generation of expression cell banks is feasible within one week while generation of stable protein production cell line takes 6-8 months.
